Selank is a synthetic heptapeptide derived from tuftsin with reported anxiolytic properties via non-benzodiazepine pathways. Human data are limited to Russian clinical studies in generalized anxiety disorder (GAD) and related conditions.

## Human Clinical Evidence

Two randomized studies compared Selank to benzodiazepines in patients with anxiety disorders.

In one trial of 62 patients with GAD or neurasthenia, Selank produced anxiolytic effects comparable to medazepam with additional nootropic benefits and no sedation or dependence noted.

A second study of 60 patients with phobic-anxiety and somatoform disorders found Selank had pronounced anxiolytic and mild nootropic effects lasting one week post-treatment, with positive impact on quality of life.

No human trials specifically examine Selank as adjunct to SSRIs for residual anxiety.

## Preclinical and Mechanistic Data

Selank exhibits concentration-dependent allosteric modulation of GABA receptors, distinct from classical benzodiazepine binding sites.

Animal models demonstrate anxiolytic activity without sedation typical of benzodiazepines; effects include gene expression changes in neurotransmission pathways.
